Hepatoprotective activity against acetaminophen-induced liver dysfunction and GC-MS profiling of a brown algae Sargassum ilicifolium

Abstract Background Drug-induced hepatotoxicity is one of the most important causes liver dysfunction. Acetaminophen (paracetamol) an analgesic-antipyretic drug generally considered safe but its overdose may cause toxicity. Marine macro-algae (seaweeds) especially brown seaweeds possess unique biological activities including hepatoprotective potential. The current study focused on effect different solvent fractions Sargassum ilicifolium and characterization n -hexane soluble fraction. Methods ethanol extract (20 g) S. was mixed with solvents increasing polarity, starting followed by chloroform methanol. All three ( -hexane, methanol) were administered to rats at dose 150 mg/kg, b.w. Intraperitoneal administration acetaminophen (600 mg/kg b.w.) used injury. hepatic damage evaluated markers enzymes; aspartate aminotransferases (AST), alanine (ALT), alkaline phosphatase (ALP), lactate dehydrogenase (LDH), bilirubin along other metabolites i.e., triglycerides, cholesterol, urea, glucose creatinine. Lipid peroxidation glutathione estimated in tissue. n- Hexane fraction subjected GC-MS analysis order identify potent compounds. Results oral methanol reduced acetaminophen-augmented marker enzymes ALT, AST, ALP, LDH, bilirubin, creatinine, triglycerides. also improved antioxidant level via enhancing reversing lipid peroxidation. spectroscopy revealed presence some new Among them, fatty acids found be highest concentration halogenated hydrocarbons, benzene derivatives, sterols. Fatty acid seaweed factors for hepatoprotection from drug-induced hepatotoxicity. Conclusion From results, it evident that have ability protect against toxicity, which comparable silymarin as a standard drug. contains bioactive compounds pharmaceutical importance.
